I am using draw.io to make diagrams related to genetics. It is convention in my field to italicize gene names.
I would like to have a box that says "genename::cDNA" where "genename" is italic and "cDNA" is not. Is this possible?

Select any/all edges that have the wrong rounding. On the right panel, under the style tab, there's a pull-down to select the edging stlye:
Select "Sharp" in that pull-down. Further down that panel is a "Set as default style" button that will set the current style for all new edges.
draw.io doesn't currently have the capability to have mutliple, separate diagrams within one file. There is a feature request open for this item.

A bit late now, and maybe obvious, but draw.io now supports the "Formatted Text" Text option which allows you to apply bold, italics, etc to any text block or sub-regions of it.
